In first part of this study,we analyzed the relationship between MGMT protein and its prognostic significance in breast carcinoma. Methods The expression of MGMT protein in 135 cases of breast carcinoma was detected by immunohistochemical method,and exploring the possible functions of MGMT in breast carcinoma development and progression. Results The expression rates of MGMT protein was 84.4%(114/135), while 21 patients had no detectable or low level of MGMT expression(15.6%). The expression of MGMT protein had no relationship with age, menses stage, tumor size, lymph node metastasis, clinical stage and survival time of breast carcinoma(P>0.05),but had close relationship with the ERαexpression(P<0.025). Conclusion The expression of MGMT protein can not be a prognostic factor of breast carcinoma.In second part, we investigated the expression of MGMT protein in breast carcinoma and the sensitivity of tumor cells to alkylating drugs. Method The expression of MGMT protein in 115 cases of breast carcinoma were detected by immunohistochemical method and the relationship between MGMT protein expression and the effect treated with CTX and/or DDP as well as PTX or TXT were studied. Results The high expression rates of MGMT protein in 115 cases observed was 86.1%(99/115). There seemed no correlation between MGMT expression and the tumor sensitivity to PTX or TXT (P>0.5),but were closely correlated with the sensitivity of alkylating agents treatments(P<0.01). Conclusion Detecting the expression levels of MGMT protein in breast carcinoma has important values for predicting the therapeutic effect of alkylating drugs and for guiding individual clinical chemotherapy.
