Osteoporosis is a metabolism disease of micro bone tissue disorganization , bony mineral matter and bony matrix reduce at same rate, sclerotin turn week, trabecula of bone decrease ,which make the brittleness of bone raise and fracture easily.It is common illness and frequently occurring illness in old people ,even in menopause women.Now ,people have no enough knowledge about the characteristic of osteoporotic fracture healing,and neglect the characteristic of osteoporosis that result in bad healing ,even fracture again.Objective: To observe the effect of transform growth factor-β( TGF-β) and insulin-like growth factor-I( IGF-1) by assess the creation and changes of expression of TGF and IGF-I in defferent period of the osteoporotic facture healing.Methods:90 wistar female rats of 250~300g and 6~9months old were randomized into two groups.One was ovariectomied group(OVX) of 60, the other was control group(sham) of 30;Creat osteoporotic model and fracture model. When at 1,7,14,2l,28,and 35th day,Two groups then divide 6 groups at random , OP group had 10 each rats ,CP had 5 rats each. Fixation , Decalcification, embedding , cut sheet ,then assess the creation and changes of expression of TGF-βand IGF-I in fracture cellus by immunohistochemistryResult: 1. During the healing process, endochondral bone formation delayed, while bony callus tissue remodeling, bone resorption accelerated . And the bone trabecula were loose.2.Without deference ,TGF-βexpressed in the cartilage cell , osteoblast and mesenchymal celland, the peak emerged endochondral bone formation(21day)(p<0.05). Compared with contral group (21day) ,the expression reduced significantly. (p<0.05). Compared with contral group (other days),the expression reduced unsignificantly. (p>0.05).3. Without deference , IGF -1 expressed in the cartilage cell , osteoblast and mesenchymal celland, the peak emerged endochondral bone formation (14 and 21 day) (p<0.05) .Compared with contral group (21day) ,the expression increased significantly. (p<0.05). Compared with contral group (other days),the expression increased unsignificantly. (p>0.05).Conclusion: 1. During the healing process, endochondral bone formation delayed and the qualities of bone cellus are adversed.2. TGF-βand IGF-1 acted as an important role in every stage of fracture healing, the change significantly in the endochondral bone may delay the endochondral bone formatted.
