| Purpose:The purpose of this study was to profile tissue specific gene expression in colorectal cancer to identify the different expressed genes in colorectal Adenocarcinoma, Adenoma and Normal Tissue and to find new targets and strategies for the management of this disease.Experimental Design:Affymetrix oligonucleotide microarrays HG-U133 plus2.0 were used to monitor gene expression of 47,000 transcripts and variants to detect differences in gene expression in human colorectal adenocarcinoma, adenoma and normal tissues. To identify genes that are important in regulating the growth properties of colorectal cancer,CRC-related genes were functionally classified by utilizing the bioinformatics method.Results:1,2340 of CRC-related genes & ESTs were obtained: 609 genes were commonly up-regulated in adenocarcinoma , whereas 1731 genes were commonly down-regulated in adenocarcinoma2,726 of early CRC-related genes & ESTs were obtained:133 genes were commonly up-regulated in both adenoma and adenocarcinoma, whereas 593 genes were commonly down-regulated in both phases3,Utilizing the software of"David",CRC-related genes and early CRC-related genes were functionally classified. Their molecular functions and biological process were known. The functions of CRC-related genes include:cell growth and /or maintenance, response to external stimulus, signal transduction, immune response, protein metabolism, nucleobase, nucleoside, nucleotide and nucleic acid metabolism and so on.The functions of early CRC-related genes include:cell growth and /or maintenance, signal transduction, protein metabolism, response to external stimulus, nucleobase, nucleoside, nucleotide and nucleic acid metabolism, biosynthesis and so on.Cell growth and/or maintenance is the biggest one in all the function groups in CRC-related... |
