Expression Of Intercellular Adhesion Molecule-1 (ICAM-1) And Lymphocyte Function-associated Antigen-1(LFA-1) In Salivary Glands Of Patients With Sjogren's Syndrome

Objective: We investigated the expression of ICAM -1 and LFA-1 in minor salivary glands of patients with SS in order to explore the role of them in pathogenesis and provide a theoretical basis for clinical therapy.Methods: We studied 30 untreated and recently diagnosed patients with primary SS and 21 healthy controls. Salivary gland biopsies were assessed for lymphocyte focus score and expression of intercellular adhesion molecule-1, lymphocyte function-associated antigen-1. Anti-Ro and anti-La antibodies were measured. Semiquantitative analysis was performed by image analysis software.Results:(1)In salivary gland samples, the high expression of both ICAM-1 and LFA-1 was significant compared to controls.(p<0.01) (2)There was a positive correlation between lymphocyte focus score and ICAM-1 expression(p<0.01) as well as LFA-1.(3)We detected linear correlation between expression of ICAM-1 and LFA-1 .The correlation coefficient was r=0.989,(p<0.01).linear regression equation is Y=1.037X+4.612×10-4(4)ICAM-1 expression was detected in 38% of ductal epithelial cell of SS patients ,and ICAM-1-expressing glandular epithelial, myoepithelial, endothelial, et al were observed throughout glandular tissues. While LFA-1 expression was detected majorly in infiltrates.(5)The percentage of anti-SSA antibodies positivist was 67% ,and anti-SSB was 47%. The expression of LFA-1 in the antibody positivity patients was significant compared to the antibody negtivity patients(p<0.01).(6)There was correlation between the constitutional symptom and anti-SSA and anti-SSB.The value of x2 are 12.129and15.200 (p<0.005)Conclusion:The ICAM-1/LFA-1 pathway may have an importent role in the pathogenesy of SS. The high expression of ICAM-1 in the salivary gland epithelial cells indicats that they act as nonprofessional APC,these cells actively paticipate in the induction and maintenance of lymphocytic infiltrates of patients. Anti-Ro/SSA and anti-La/SSB antibodies are important index of SS,the relationship between antibodies and LFA-1 suggested activation of B lymphocytes relate to lymphocytic infil -tration .The emergence of constitutional symptom related to antibodies positivity .
