Part 1 : A screening assay for the Hypsi-invasive subset in vitro from the human invasive carcinoma cell strain of urinary bladder,T24Objective: To get the subset with hypsi-invasiveness form the human invasive bladder transitional carcinoma cell strain,T24, and to observe the changes of invasive capability and motility.Methods: Improved Boyden chamber system was utilizated to obtain hypsi-invasive subset, named T24-Ⅲtemporarily, from human bladder transitional carcinoma cell strain, T24, after a serial passage in vitro. The differences of the growth curves, invasive capability and motility between the couple of cells were evaluated by the same test facilities. The expression of MMP-7mRNA was detected by fluorescent quantitation of reverse transcription-polymerase chain reaction.Results: Daughter cells named T24-Ⅲwas obtained, whose morphologic characteristics, status and required cultural circumstance were no obvious variance when being compared with T24. Before the 7th day during cell culture, there was no marked change of their growth activities between the offspring and parent cell strains. But at the 8th days, T24-Ⅲwas a little more proliferative than T24 in the same cultural conditions. The mean of whole invasive and chemotaxis motor cell populations of T24-Ⅲwere T24's 3.4 and 3.2 times respectively, which manifests that T24-Ⅲis more invasive and locomotor than the parent cell strain T24 in vitro(p<0.001). Furthermore, the invasive power of the two groups of bladder cancer cells was positively correlated with their chemotaxis motor abilities, that the standardized contiguous coefficient was 0.858 (p<0.001). The MMP-7mRNA numerus of T24-Ⅲand T24 were 22.8828±24.2435 and 12.4750±11.2905. There might be a difference at the expression of MMP-7mRNA between T24-Ⅲand T24, however, it was not statistically significant with Wilcoxon rank sum test(p=0.08).Conclusion: Firstly, The hypsi-invasive subset, named T24-Ⅲ, was obtained successfully from human bladder transitional carcinoma cell strain, T24, using improved Boyden chamber. The invasive power and chemotaxis motor ability of T24-Ⅲare more powerful than T24 in vitro.Secondly, the invasiveness of the two groups of bladder cancer cells is positively correlated with their chemotaxis motor abilities. Thirdly, the expression of MMP-7mRNA of T24-Ⅲstepped up tendencily comparing with T24. But it was not statistically significant.
