Objective:Hepatocellular carcinoma (HCC) is one of the most common malignant tumors in our country, which is frequently associated with high lever of malignancy, poor prognosis and recurrence after operation. It is beacause of the invasion and metastasis of HCC. To identify the mechanisms of invasion and metastasis and to search effective approaches to prevent the recurrence and metastasis of HCC remain a challenge to modern medical science. Studies indicate that there are two major mechanisms of invasion and metastasis in hepatocarcinoma: one is tumor cell penetrating extracellular matrix (ECM) and vascular basement membrane and the other is angiogenesis. Heparanase (hpa) is an endo-β-D-glucuronidase and it is the only one we know by now that degrades the heparan sulphate chain of heparan sulphate proteoglycans (HSPGs), and then degrades the extracellular matrix and basement membrane, releases and activates growth factors combined with HSPGs such as bFGF, VEGF et al, and induces angiogenesis, in this way, Hpa promotes tumor invasion and metastasis. The mechanisms of Hpa regulation are not clear, but it may be associated with hypoxia in tumor tissue theoretically. The transcription factor hypoxia inducible factor 1 (HIF-1) is a...
