Objective: In order to discuss the role of NO in visual development and deprived amblyopia, we used nNOS antibody to observe the expression and variation of nNOS in dLGN and visual cortex 17 area of the rats with monocular deprivation before and after reverse suture treatment, then approached the changes of neural activity level in visual center, providing the molecular theory basis and a new way for the prevention and treatment of amblyopia.Methods1, Establishment of animal model and group division: we sutured the eyelids of one eye of 14-day-old male rats. Two weeks later, some of them were opened deviated eyes and occluded non- deviated eyes for reverse suture treatment. The rats were randomly divided into three groups: (l)Monocular deprivation( MD) group(12 rats), their left eyelids were sutured at postnatal day 14, then were further divided into two subgroup: MD( I )and (II), respectively sacrificed at two and four weeks after operation(postnatal day 28 and 42). (2)Reverse suture(RS) group(6 rats), they were sacrificed at postnatal day 42. (3)12 normal rats were used as controls(NC group), they were divided... |
