Objective To explore the dose and time correlation between the expression of connexin43 gene, the most common form of the connexins ,and ionizing radiation(IR),one of the carcinogens; and also research that wether connexin43 could be a new valuable biomarker for the estimation of ionizing radiation damage or not.Methods We established the model of human normal lymphoblastoid AHH-1 cells irradiated with different γ-ray doses, then using the cDNA microarray screened some radiation-induced genes, the changed transcriptional level was further confirmed by RT-PCR assay for some of these genes; finally we concretely analyzed the expression of radiation-induced gene connexin43 with the methods of Western blot and Northern blot.Results 1) Human normal lymphoblastoid AHH-1 cells were irradiated with cobalt-60 y-ray in 2Gy as well as 10Gy at 4h post-irradiation (un-irradiated cells as control), cDNA microarrays displayed 21 up-regulated genes and 12 down-regulated genes in 2Gy irradiated cells, while there were 299 genes down-regulated and 15 genes up-regulated in 10Gy irradiated cells.2) cDNA chip displayed 11 genes were up-regulated and nine were down-regulated, The changed transcriptional level was further confirmed by RT-PCR assay for some of these genes, i.e.CONNEXIN43, BMPR2, NOL6, LOC5176, LYK5, etc.after Human normal lymphoblastoid AHH-1 cells exposed to 5cGy γ-ray at 4h post-irradiation(un-irradiated cells as control).3) Western blot and northern blot were used to explore the expression level of connexin43 gene after exposed to different dose irradiation. The results demonstrated connexin43 gene has been remarkedly up-regulated and that there would be dose and time correlation between the expression of connexin43 gene and γ-rays.
