| Objective:To study the change rules of the epidermal stem cells during the process after full-thickness skin autograft on rat. These results provided a theoretical evidence for the fundamental investigation to clinical application; and a theoretical basis for improving the quality of full-thickness skin autograft in practices. It's also a base for further study of the tissue engineering skin.Methods:Forty-two wistar rats, which were two to three months old, were randomly divided into seven groups: group 1 (one day after operation), group 2 (three days after operation) , group 3 (five days after operation) , group 4 (seven days after operation) , group 5 (fourteen days after operation) , group 6 (twenty-one days after operation) , group 7 (one month after operation). Each rat was made full-thickness skin autograft model on back about 1. 5cm x 1. 5cm square. At the operating time and the different 7 times after operation, samples were harvested from each rat in each group. By the method of S-P immunohistochemistry, integrin β1 and P63 were employed as the first antibody to demonstrate the epidermal stem cells. Each immunohistochemistry sample was observed under two hundred multiples optical microscope; And each sample was observed five different scans; And count one hundred epidermal cells and positive cells in each scan; Then computed the positive cells ratio (PCR, %). The results were compared between before and after operation in each group, also among the all groups after operation. |
