And the specimens of normal tissues were taken from the resected colorectal spicemens above 10cm far from cancer. Four micrometers thick sections prepared from formalin-fixed and paraffin-embedded tumor tissue or normal tissue on the slides were stained immunohistochemically with the primary antibodies as mouse monoclonal anti-human E-cadherin orβ-catenin (Fu Zhou Maxim Biotechnology, ready to use) and the SP kit(Fu Zhou Maxim Biotechnology) . Nomal mucosal epithelium served as an internal positive control and adjacent normal mucosal epithelium as an internal positive control . Negative control was obtained by replacing the primary antibody with PBS. Based of the previous immunhistochemical studies,the membranous staining of cells were positively stained with brown by the antibody. Calculating the percentage of positively membranous stained cells evaluated for each tissue section after counting 100 cells at more than 5 high-power(×400) fields.
