Objective: To investigate the effect of an antioxidant edaravone on the anti-apoptotic and neuroprotection in a focal ischemic model in rats.Methods: The model of focal cerebral ischemia in rats was established by Suture occlusion method. Rats were randomly divided into sham-operated group, reperfusion group and edaravone group. Reperfusion group and edaravone gruop were subdivided into 2-, 6-, 12-, 24- and 48-hour groups according to reperfusion time. Rats in edaravone group received edaravone (3mg/Kg) intravenously administered after occlusion for 2 hours, and the same dose of edaravone was used again after the primary administration. Rats in reperfusion group received physiologic saline with the same dose, method, time as the edaravone group. The malondialdehydc (MDA) concentration of Blood serum was determined by colorimetric method. Bcl-2 immunohistochemistry and TUNEL staining were performed to determine the expression of the Bcl-2 protein and number of TUNEL-positive apoptotic cells in brain tissue. Infarction volumes were assessed as well.Results: The volumes of infarction in the edaravone group were obviously smaller as compared with that of reperfusion group after reperfusion for 6, 12, 24, 48 hours (P<0.05). MDA concentration of serum in the edaravone group was significantly lower than that of the reperfusion group after reperfusion for 6, 12, 24, 48 hours(P<0.05).
