| Objective 1. To probe the relationship between CTGF and diabetic nephropathy in diabetic patients and experimental diabetic rats;2.To explore the effects of Rosiglitazone and Simvastatin on renal CTGF expression and rxcretion in diabetic rats and their renoprotective mechanisms.Method 1. Sixty patients with diabetes were divided into three groups according to urinary albumin excretion rate(UAER) : normal albuminuric group (A) ( <20μ g/ min ,N = 20) ,microalbuminuric group (B) (20-200 μ g/ min , N = 20) and macroalbuminuric group (C) ( >200 μ g/ min ,N = 20) . The urinary CCN2 was determined by ELISA. 2. Four groups of rats were studied: group A (n=8),normal control rats; group B (n=8), strephtozotocin induced diabetic rats; group C (n=8),diabetic rats treated with Simvastatin (20mg/kg.d); group D (n=8), diabetic rats treated with Rosiglitazone (4mg/kg.d).Blood glucose(by glucose oxidase) ,CTGF(by ELISA) were measured at the lst,4nd,6th and 8th week ,as well as the expressions of CTGF protein (by histochemical staining) and the relative kidney (ratio between kidney weight and body weight) at the 8th week. Resμlt 1.(1) The excretion rate of the urinary CCN2 in diabetic patients was markedly higher than that in healthy controls( P < 0. 05) moreover ,the excretion rate of urinary CCN2 was increased with UAER and there were significant differences among the three groups( P < 0. 01) .Urinary CCN2 was also positively correlated with UAER and RBP ,negative correlated with creatinine clearance ( Ccr) 2 .(1)At the 1st week(no treatment given), the urinary excretion rates of CTGF in group B, C and D were significantly higher than that of group A (P<0.01).
