OBJECTIVE: Cerebral ischemia-reperfusion injury (CIRI) triggers a multitude of pathophysiological and biochemical events. Therefore, the treatment of it should logically employ some known neuroprotective agents because only using one neuroprotective drug can not block the evolution of CIRI completely. We hypothesized that a treatment combining danshensu and hydrosafflow yellow A might have a potential neuroprotective effect. To assess this idea, a rat model of transient focal cerebral ischemia-reperfusion was established. Neurological deficits score and infarct volume and pathological examination were used to evaluate the synergistic effect of combining danshensu and hydrosafflow yellow A. Subsequently, to investigate the mechanism of combined danshensu and hydrosafflow yellow A , we measured neural cell apoptosis in the hippocampus of ischemia hemisphere and examined the mRNA and protein expression of bcl-2,bax and Caspase-3 after focal cerebral ischemia-reperfusion injury in rats.METHODS: The models of focal brain reperfusion injury by middle cerebral artery occlusion(MCAO)were established in adult wistar rats weighing 270-330 g. 200 rats were randomly divided into 8 groups: group I (sham), anesthesia and NS intravenously (iv) only; group II ,ischemia-reperfusion(IR)and NS iv;...
