[Objective]This study aimed to observe the mechanism of hepatic Ischemie-Reperfusion and the effects of propofol in rat in vivo.[Methods] Firstly, to build the model of hepatic Ischemie-Reperfusion in rat according to the method of Kobayashi, and then seventy-two Wistar rats weighing 250350g were anesthetized with injection by intraperitoneal 3% pentobarbital 30 mg·kg-1'.The animals were randomly divided into the control group(group A),ischemia and reperfusion group(group B),I/R+propofol 4 mg·100g-1·h-1 (group C), I/R+ 6mg·100g-1·h-1 group(group D).Each group divided into four subgroups(n=6). Blood samples and liver tissue were obtained at 30min-ischemain and 30min-reperfusion (T1), at 60min-ischemain and 60min(T2), at 60min-ischemain and 120min-reperfusion(T3). The serum levels of TNF-α and IL-10 were measured with ELISA.The activity of NF-kB were detected with EMSA.The ultramicrostructure and cell apoptosis and were observed by electron microscope.[Results] 1. The serum levels of TNF-a at T1 and T2 and T3 in group B were significantly increased as compared with the group A(P<0.05) , To campared with group B,the levels of TNF-a were decreased but the levels of IL-10 increased significantly(.P<0.05)in groups of C and D. However,the levels of TNF-a and IL-10 were no significant difference between group C and D(P>0.05).
