| Bombyx Batryticatus is a commonly used Chinese medicinal materials, which are the larva in 4th to 5th stadium of Bombyx mori Linnaeus infected by Beauveria bassiana (Bals.) Vuillant. Bombyx Batryticatus mainly comes from Sichuan, Jiangsu, Zhejiang, Guangdong and Shandong provinces, while Sichuan has its largest production. Bombyx Batryticatus has abirritative, relieving a cough, detumescent and defervescence. It has been used for the treatment of eclampsia, epilepsy, mumps, fauces ache, upper respiratory infection, enuresis and face nerve numbness. Chinese Pharmacopoeia ediction 2005 describes Bombyx Batryticatus's source, exterior character, microstructure, physical and chemic character. Obviously, the ingredients indicate Bombyx Batryticatus's main efficacies is ambiguity, which present quality standard can't estimate its internal quality exactly. In order to improve the quality standard of Bombyx Batryticatus, we have undertaken a research on sieving representative ingredients and constituting internal quality standard of Bombyx batryticatus.Firstly, we have compare the pharmacological effect of different parts of Bombyx Batryticatus. According to its main efficacies, we separated the extracts of Bombyx Batryticatus into petroleum ether extracts, chloroform extracts, ethyl acetate extracts, water extracts and extracts which had been extracted by ethanol. Nitrostrychnine, nikethamide and isoniaxide convulsions were respectively induced by hypodermic injection of nitrostrychnine, nikethamide and isoniaxide. As a result, we found that the anticonvulsant active components exist in chloroform and ethyl acetate extracts.Secondly, we have completed the systematic chemical studies on Bombyx Batryticatus, by using the methods of Macroporous adsorption resin, Silical gel column chromatography to separate and purify the chloroform extracts. Five compounds were isolated and identified as 6,9-epoxy- ergosta-7,22-dien-3-ol, β-sitosterol, ergost-6,22-dien-3β. 5α. 8α-triol, beauvericin, β-daucosterol by MS, 1H-NMR, 13C-NMR. 6,9-epoxy-ergosta-7,22-dien-3-ol were isolated from Bombyx Batryticatus for the first time. We have researched the anticonvulsant active components by hypodermic injection of nitrostrychnine, nikethamide and isoniaxide, for the first time, we found beauvericin and ergost-6,22-dien-3β, 5α, 8α-triol are the anticonvulsant active components. We also have established the quality standard of Bombyx Batryticatus and completed the instruction. We took beauvericin as reference substance to setup the TLC method, RP-HPLC method of Bombyx Batryticatus, for the first time. The second method is convenient and sensitive, and the sample application recovery rate of beauvericin is 99.19% and RSD is 1.40%, which can be used as the index of quality control. We have determined sixteen type of sample from different habitat by using this method, the total content of beauvericin arranged from 0.0316mg/g to0.6259mg/g. Furthermore, we have mensurated the content of inorganic elements in different Bombyx batryticatus. As a result, Bombyx Batryticatus have high content of magnesium. Meanwhile, the content of lead is more than 3 times than that in Chinese Pharmacopoeia ediction 2005. Further research on how to control the content of heavy metal in Bombyx Batryticatus should be well developed. |
