Studies On The Constituents From Arnebia Euchroma (Royle) Johnst And Lithospermum Erythrorhizon Sieb. Et Zucc And The Preparation Of Zicao Scalding Ointment

Naphthoquinone pigment is the main constituents of plant Zicao. The structure of 5, 8-dihydroxy- naphthoquinone makes it instable with light and heat. The effective compounds are easy destroyed by means of solvent extracting. In the research, CO₂ supercritical fluid extraction was used to extract the naphthoquinone pigment from the dry roots of Arnebia euchroma (Royle) Johnst and Lithospermum erythrorhizon Sieb. et Zucc at high pressure and normal atmospheric temperature.By means of the silica gel chromatography, 10 compounds were isolated with the elution of petroleum ether-CHCl₃, petroleum ether-EtOAc and cyclohexane-CHCl₃ system. On the basis of physicochemical properties and analysis of UV, IR, NMR data, the structures of them were identified. Among them, 4 compounds of deoxyshikonin, β,β-dimethylacrylshikonin, acetylshikonin, shikonin were isolated from Armbia euchroma (Royle) Johnst; the others, such as deoxyshikonin, β,β-dimethylacrylshikonin, isobutyrylshikonin, acetylshikonin, shikonin and β-sitosterol were isolated from Lithospermum erythrorhizon Sieb. et Zucc.
