Lithospermum erythrorhizon Sieb.et Zucc is mainly distributes in Northeast with large amount. It is showed that the Naphthoquinone pigment which have the structure of 5,8-dihydrohy- naphthoquinone is the main constituents of plant Zicao.First, CO2 supercritical fluid extraction was used to extract the naphthoquinone pigment from the dry root of Lithospermum erythrorhizon Sieb.et Zucc at high pressure and the normal temperature. By means of Si-gel cc and demipreperation-HPLC, five compouds(CS1~CS5)have been gained with the elution of petroleum ether-EtOAc system. Though the physicochemical properties and analysis of UV,IR,MS,NMR data ,we were known those chemical structures: isobutylshikonin , isovalerylshikonin, acetylshikonin,β-hydroxy-isovalerylshikonin ,shikonin.Secondly, a RP-HPLC method for simultaneous content determination of six compounds such as isobutylshikonin , shikonin, acetylshikonin, isovalerylshikonin,β-hydroxy-isovalerylshikonin,β,β-dimethylacrylshikonin in Lithospermum erythrorhizon Sieb.et Zucc was established. The method displayed excellent linearity, precision, reproducibility and stability. It was showed that the method was simple, accurate and reproducible, which provided a basis for application in assaying studies and routine determination of numerous samples of plant Zicao.Finally, with the method mentioned above, the samples from 10 producing areas in Neimenggu and Heilongjiang were detectded. It was showed that the quality of Yingzicao in Xiawazi was best.
