ObjectiveTo explore the mechanism of Bushen-Yiqi-Huayu prescription which ameliorates the learning-memory ability of subacut aging rats, by observing the change of rats' behavior with water maze test and hippocampi's micro-structure through microscope,examining the change of enzymology in hippocampi with ultraviolet spectrophotometer. MethodsSixty wistar rats which were 3-4 moon's age were adjustably bred for 1 week,6 rats fell into disuse for out of the 90% region by morris water maze test ,the rest 54 rats were divided into 5 groups:blank group(10 rats), model group(11 rats). drug(low,middle, high)groups(11 rats each).The model was set up by injected with 1% D-galactose 50mg/kg into abdominal cavity for 6 weeks in model and drug groups,but blank group with saline.drug(low, middle, high) groups were administrated with extrated fluid of Bushen-Yiqi-Huayu prescription (4g/kg 8g/kg 12g/kg) for 4 weeks,blank and model groups with saline.The learning-memory ability of rats were tested by morrise water maze , recorded the types of track and latent period; The rats were texecuted after experiment, detecting the content of MDA and enzymology activity of SOD,AchE in hippocampi of rats by ultraviolet spectrophotometer,observing the hippocampi's micro-structure through microscope. ResultsTo blank grouprthe times and percentage of correct searching of rats in water maze were 64 and 88.9%,the average latent period in after experiment[ (12.30 ± 2.95 ) s] had less time than before [ (18.05 ± 3.92 ) s] (p < 0.01 ); the each of the content of MDA and enzymology activity of SOD,AchE in hippocampi was (3.58 ±0.90)nmol/mgprot, (515.19 ± 55.27)U/mgprot, (0.39 ± 0.04)U/mgprot; the picture's color was dark under microscope with clear outline ,the closely packing cells which had more branches , big deep-dyed nucleus and clear nucleolus sublobe arranged orderly with distinctness;there were plenty cell organs , clear mitochondrion's crest on which had much ribosome's grains and big nucleus which had no-crimpling nuclear membranes , clear nucleopores and large painting-uniform nucleolus and rich synapses which had normal gaps in the neuron of CA3 zone in hippocampi under the electron microscope.To model group: the times and percentage of correct searching of rats in water maze were 35 and 48.9%, the average latent period in after experiment[ (32.25 ± 9.57 ) s ] had more time than before [ ( 18.06 ± 2.91 ) s] ( p < 0.01 ); the each of the content of MDA and enzymology activity of SOD,AchE in hippocampi was (3.50 ± 1.06)nmol/mgprot, (404.61 ± 66.90)U/mgprot, (0.50 ± 0.04)U/mgprot; the picture's color was light under microscope with blurry outline ,the swoln-denatured cells which had less branches and small concentrated nucleolus with unclear sublobes arranged loosely each other;The dropsical cells contained little cell...
