Objective: To make isolation of chemical constituents from'Banlangen', root of Isatis indigotica Fort.(Cruciferae), and study their immunocom- petence screening.Methods:1. Making isolation with dry-column flash chromatography, classic silica Chromatography and activated charcoal dry-column flash chromatogra-mphy. 2.Elucidation of chemical structure of compounds with 1HNMR, IR, 13CNMR, MS.3. Immunocompetence screening of constituents from Isatis tinctoria Fort. in vitro: the immunoregulation of the fractions and compound A. were detected by proliferation of mice splenocytes (MTT assay) and mixture lymphocyte reaction (MTT assay).Results:1. Fractions and 8 compounds were isolated from 'Banlangen', root of Isatis tinctoria Fort. 6compounds were identified as: icosane, heptadecanoic acid,β-sitosterol, indirubin, malonic acid, D-(+)-sucrose, the other com-pounds are stillcould not be identified; Fractions and 1 compounds were isolated from 'Daqingye', leaf of Isatis tinctoria Fort.identified as n-docosane. 2. Icosane,heptadecanoic acid, malonic acid were isolated from 'Banlangen' for the first time. n-docosane was isolated from 'Daqingye' for the first time.3.Activated charcoal as adsorbent, improved dry-column flash chromatography .4. A two-way immunoregulation tendency was found from Banlangen-fractions and compound A.
