| Objective:This research was to explore the treatment effect of isatis indigotica fort to androgen alopecia model role, and provide experimental basis for treatment of androgenetic alopecia. The research targeted with androgenetic alopecia on mice model, topically wipe the fresh juice of isatis indigotica fort on the depilation of hair loss model with test the ratio of serum T.Methods: Weighed the 56 male B6CBAF1 / J mice in turn, numbered the mice according to the weight from small to large. Randomly choose 13 mice as blank control group according to random number table method. The rest were fed in a cage.To build the model of androgen alopecia, we take the continuous use of subcutaneous injection of testosterone propionate according to the 5 mg/kg · d dose in back skin for5 weeks, except the blank group. After 5 weeks, randomly choose and killed 3 mice from the blank control group and ungrouped mice by cervical dislocation, and its hair were observed and pathological changes in hair loss area skin tissue were researched by microscopical technique. Another 40 mice were divided into 4 groups randomly,10 mice model group, saline group,positive control group, isatis indigotica fort group.Continue to give the same dose of testosterone propionate injection, at the same time,give isatis indigotica fort group fresh juice of uxmalgrass for external use, give the positive control group 3% minoxidil solution for external use, give the saline group saline for external use, no drugs to model group. The test would be ended in 9 weeks,mice will be killed after take serum from femoral artery blood by cervical dislocation.Take back hair removal HE staining for the areas of skin tissue in mice. Calculate the proportion of the terminal hair/vellus hair. Test the serum T with Testosterone Test Kits. Analyze data in SPSS19.0.Results:1. In the process of building the animal model, the hair on the back of mice began to fade on second week, loss on 5th week and expanding, it indicate that we got successful AGA modeling from the histopathology that showed Hair follicles miniaturization the and the decreasing of the proportion of the terminal hair.2.In the 9th week, took the skin tissue of mice’ back for HE staining and count the number of terminal hair and vellus hair. It showed that the proportion of the terminal hair/vellus hair of ucmal grass group was significantly increased compared with model group and saline group(P < 0.01),decreased compared with blank control group(P<0.05),no statistical difference compared with positiVe control group. 3.When the experiment ended in 9th week, we made a comparison between groups of mice serum content of T. Compared with the black group, serum content of T of other4 groups was significantly increased(P<0.01), and there is no significantly difference in intraclass of the content of serum levels of T(P>0.05).Conclusions:1. B6CBAF1 type/J mouse is a kind of ideal model to build the model of androgen alopecia stablely.2.The treatment of isatis indigotica fort for AGA were effective and had no statistical difference compared with positive control group.Isatis indigotica fort can improve Hair follicles miniaturization and the proportion of the terminal hair/vellus hair.3. There is no significantly influence on T in the process of the treatment. |
