| Object:secondary infection is the predominant cause of death on the ground of necrosis pancreatitis,which is correlated with bacterial translocation closely.It is common recognize that there is IETM at the state of ANP. Bacterial translocation and IETM are correlated with intestine membrane permeability closely.So it is very important to study the change and mechanism of intestine membrane permeability at the condition of ANP Materials and methods: Healthy female sex rats 50 merely, body weight is 210-230gramme, devided into 3 groups:Sham operation SO,open abdomen, only turn over pancreas lightly, not given any treatment; Acute necrosis pancreatitis ANP, according to classic model prepared; Fasting diet FD, parenteral nutrition.At the point of 72h, killed the rats,remain blood plasm,serum,pancreatic tissue and intestines tissues pecimen, using biochemistry, radioimmunity, pathology and immunohisto- chemistry to detect correlated index. Determine endotoxin in blood plasma to assess intestinal endotoxemia; using the method of radioimmunity to detect TNF-αand IL-4 in serum to assess immune function of organism; use spectrophotometric method to determine DAO in plasma and intestinal homogenate to observe damage condition of intestine.Using iodine and starch chromatometry to determine serum amylase in serum to observe the severity of pancreatitic; intravenous inject Evans blue through vena caudalis to observe intestine mucous membrane permeability and vasopermeability;use the method of thio- pentobarbital sodium to determine malonaldehyde of serum;using fluorescence photometry to determine histamine in plasma and intestinal homogenate.HE stained display the histomorphology in intestine and pancreatic tissue; using situ apoptosis method to assess the apoptosis of enterocyte and pancreatic acinar cell.Result:ET,DAO,TNF-α,IL-4 and histamine exist significant difference among the three groups,in ANP group the level is the highest; serum AMY in ANP group has significant difference compareing with SO and FD groups; endotoxin in plasma has dependability compareing with histamine and DAO in plasma and intestinal homogenate,and TNF-αand IL-4.HE stained display the damage of intestine mucous membrane aggravated gradually, the method of situ apoptosis detect its result is coincident with the result of HE, Apoptosis cell in ANP group is the most obviously, ;pancreas patho change in SO group is approaching with normal tissue,the lesion is grave in ANP group, pancreatic tissue situ apoptosis detection is... |
