Purpose: To observe whether the leptin and leptin receptor (including ob-Ra and ob-Rb) is elevated in rat experimental kidney fibrosis induced by UUO.To observe the location of leptin receptor in the kidney of Wistar rats and analyze their role in pathogenesis of renal fibrosis. Methods:40 male Wistar rats were randomly divided into the sham operation group (n=8),day 3 after UUO(unilateral ureteral obstruction) group (n=8),day 7 after UUO group(n=8), day 14 after UUO group(n=8) and day 21 after UUO group(n=8).Rats were sacrificed at the exactly time. The level serum leptin at each time point were examined by ELISA .Leptin receptor,s location was detected in every group by immunohistochemistry. The ob-Ra and ob-Rb mRNA transcription was assessed by semi-quantity reverse transcription polymerase chain reaction (RT-PCR).Results: The level of serum leptin increased progressively at day 7,14 and 21 after UUO. The same results was observed in ob-Ra. The level of ob-Rb was not significantly higher in UUO groups than in sham operation group. Leptin receptor were localized in the epithelial of tubules.Conclusions: The coexistence of renal fibrosis and increased serum leptin and ob-Ra mRNA in renal were found in UUO rats.
