| Objective: To explore the role and relationship of thrombomodulin and plasminogen activator inhibitor-1 in the development of carotid artery lesion in DM,to provide the theoretical foundation for diagnose and therapy in early stages of carotid artery lesion occurred in DM.Methods: 48 Wistar rats were divided randomly into two groups. There were 24 in normal control group(group A) and 24 in diabetes mellitus group(group B). The subjects in group B were injected intraperitoneally with dose streptozotocin (STZ,50mg/kg)while the ones in group A were injected intraperitoneally with the similar dose of citrate buffer. Six weeks, ten weeks and forteen weeks administration, there were eight rats in both groups respectively killed. The blood samples were obtained to measure the levels of TM,PAI-1,FINS and C -peptide. The early changes of the carotid artery were examined using light microscope and electric microscope. Computorized image progressing was employed to measure the carotid artery intima-media thickness(IMT). We compare these data between the two groups and among the different stages with factorial designed analysis of variance and use Linear correlation to reveal the relasionship between TM,PAI-1 and carotid artery lesion in DM.Results: 1.There was a difference in the level of fast blood glucose between group A and group B, it was significant after injected intraperitoneally with STZ and the level of FBG was higher than 16.7mmol/L in group B .2.The levels of TM and PAI-1 of group B was much higher than group A(P<0.01) they increased continually during the whole experiment.3. Compared with group A, there were significant differences in the level of FINS and C -peptide (P<0.01). The differences between the two groups had existed for the whole course of the experiment.4. After admimistrated with STZ ten weeks later, the carotid artery intima-media thickness(IMT) of group B was higher than that of group A (P<0.01), fourteen weeks later, IMT was higher than that of ten weeks.Conclusion: The increase of TM and PAI-1 occurred before visible and measurable carotid artery pathological changes. The carotid artery intima-media thickness was correlated with the TM and PAI-1.TM and PAI-1 may play a harmful role in the progression of DM carotid artery lesion. TM and PAI-1 can be used as an early figure to predict DM carotid artery lesion. |
