Objective:To investigate the beneficial effects of aldosterone receptor antagonism-spironolactone on cardiomyocyte apoptosis and correlative gene of apoptosis after acute myocardial infarction in rats. Methods:AMI model was established with ligation of anterior descending coronary in male SD rats.48 survivors were randomly assigned to AMI control group(AMI,n=6, saline 2ml ? d-1 in drinking ) and spironolactone treatment group(Spi,n=6,spironolactone 20mg ? Kg-1 ? d-1 2ml in drinking),the others established randomly sham-operated group(Sham,n=6, saline 2ml ?d-1 in drinking). The following indexes were observed at 48h,7d,14d,21d.(1) The collagen content (hydroxyproline levels) was measured by chloramine T.(2) The ratio of type I and III collagen was assessed by immunohistochemical staining. (3)The myocyte apoptosis and the expression of bcl-2 and bax protein were detected by flow cytometry.(4)The expression level of bcl-2 mRNA and bax mRNA was detected by RT-PCR.Results: 1 The effect of spironolactone on the collagen content in non-infarcted areas: Compared with sham-operated...
