| During blood donation and transfusion, ABO and RhD typing, and HIV-1, HIV-2, HBV, HCV and syphilis testing are compulsorily required in China nowadays. Usually, blood typing is tested with slide assay, while tests for those viruses entirely depend on ELISA. Those traditional assays have no in parallel, can't be simultaneously carried out, and are time, money and energy consuming. Since chip has taken the advantages of high though-put, parallel test, time and money saving and being prone to automatization, great attention has been paid to in medical laboratory science. As a new assay, microarray is ideal to be applied in blood screening: typing and diagnostics for transfusion-transmitted viruses. In this study, our aim is to design and build up a protein chip to type ABO blood groups, in order to found a basis for the preparation of a protein chip simultaneously typing and testing transfusion-transmitted viruses in parallel.Materials and Methods:Comparison of coating Ab ability of silicic acid slides and aldehyde slides1. Preparation of silicic acid modified slides and optimization of reactive condition of antigen (Ag) binding to antibody (Ab): After sodium silicate solution was dropped onto a clean slide, the slide was dried and followed by acetic acid addition, then a film of silicic acid was formed on the surface of the slide. Anti-A and–B antibodies were coated to a slide on its silicic acid part and non-modified part. The later served as negative control #1. Normal saline was also added to silicic acid part of the same slide to be regarded as negative control #2. Then the slide was placed in a humid chamber at 4℃for overnight. 2% RBC saline solutions of corresponding blood types were added to the slide. RBC number was counted with microscopy after 5 minutes'reaction and PB (pH8.0) rinsing.20 groups were carried out with a cross-mix of reaction time (3, 5, 10 min), reaction temperature (24℃and 37℃)and pH values (5.7, 6.5, 7.0, 7.5, and 8.0).
