It is well known that space flights could lead to osteopenia, which is manifested by the loss of bone mass and bone mineral, declines in bone mechanical function. These changes appear to be processive, which would destroy bone structure and function in long-term space flights. Bone mass changes are site-specific rather than evenly distributed throughout the skeleton with the tendency that weight-bearing bones are more affected than non-weight-bearing bones. Even after several decades of extensive researches, the basic mechanisms of osteopenia induced by weightlessness (OW) have not been fully elucidated yet, and the physiological effects of countermeasures against OW are still limited. Understanding the nature of the effects of weightlessness on skeleton system and developments of new effective and efficient countermeasures against osteopenia have therefore become one of the primary tasks of the space life sciences research effort.Mechanic stress is one of the most important factors in remodeling process. To adapt to the environment of structure and function of bone, bone continue remodeling according to the force on it. Osteoblast can sense many kinds of strain, including compressive stress, tensive stress, benting stress and shear stress. And osteoblast is most sensitive to shear stress. Runt-domain family include Runt of drosophila and Cbfa1, Cbfa2, Cbfa3 of mice and human being, It is a group transcription factors which have important roles in organ-formation, genesis of nervous system, development of bone and formation of hemopoiesis. Cbfal, as an osteoblast-specific transcription factors, plays a key role in skeleto-development and formation.
