Objective To investigate the difference of tumor necrosis factor-related apoptosis-inducing ligand-receptor 2 and 3 (TRAIL-R2 and TRAIL-R3) expression in human non-small cell lung cancer(NSCLC) ;To investigate the correlation of TRAIL-R2 or TRAIL-R3 expression with the histopathology, differentiation and clinical staging of NSCLC and to predict the therapeutic potential of recombinant human TRAIL (rh-TRAIL) in NSCLC in the future.Methods (1)60 tissue samples were obtained from 50 patients with NSCLC and 10 patients with various benign pulmonary diseases. The samples were re-divided into the squamous carcinoma group( (ns=32) and adenocarcinoma group (na=18) ; Of all NSCLC samples, 14 were poorly differentiated, 17 were moderately differentiated and 19 were well differentiated; Among them, 8 and 6 samples were classified into the poorly differentiated squamous and adeno carcinoma group; 10 and 7 in moderately differentiated squamous and adeno carcinoma group; 14 and 5 in well differentiated squamous and adeno carcinoma group; According to TNM classification, there are 14 stage I cases, 22 stage II cases, 12 stage III cases and 2 stage IV cases. (2)A11 the samples were analyzed for the expression of TRAIL-R2 and TRAIL -R3 using Western blot method.Results (1)The expression of TRAIL-R2 and TRAIL-R3 were both lower inmalignant NSCLC group than that in benign lesion group ( P<0.05). (2) In NSCLC group,TRAIL-R2 expression was higher than TRAIL-R3 expression , while both receptors werehighly expressed in the benign lesion group. (2)No significant correlation was detectedbetween TRAIL-R2 and TRAIL-R3 in both NSCLC and benign lesion group(P>0.05) .(3)No significant correlation was detected between TRAIL-R2 or TRAIL-R3with histopathology of NSCLC (rTRAIL-R2= -0.2413,P>0.05; rTRAIL-R3= -0.0585, P>0.05) .(4) TRAIL-R2 and TRAIL-R3 were both positively correlated with the differentiationlevel of NSCLC.
