And to explore the aberrant expression mechanism of E-cadherin-catenin complex and the correlation of E-cadherin-catenin complex with Wnt pathway. Methods: 102 cases of gastric adenocarcinoma and para-cancer mucosa were accumulated and made into three tissue microarrays which containing 282 dots, 156 dots and 156 dots respectively. Expression of E-cadherin,β-catenin, c-ErbB2 and c-myc proteins were detected by immunohistochemical staining to these tissue microarrays. Results: The effective rate of our tissue microarrays was 98.01%. In gastric adenocarcinoma, the rate of abnormal E-cadherin andβ-catenin staining was 84.0%(84/100) and 76.0%(76/100) respectively, the cytoplasmic/nuclearβ-catenin accumulationthe rate was 57.0%(57/100), the expression rate of c-ErbB2 and c-myc was 29%(29/100) and 72%(72/100) respectively. No correlations of expression of E-cadherin andβ-catenin with clinicopathological parameters but histological grade (P<0.05) and depth of invasion (P<0.05) were found. The expression of c-ErbB2 was closely relatd to histological grade(P<0.05) and depth of invasion (P<0.05). The expression of c-myc was closely relatd to histological grade. The aberrant expression ofβ-catenin was significantly correlated with the expression of E-cadherin, c-ErbB2 and c-myc in gastric adenocarcinoma(P<0.05). The abnormal expression rate of E-cadherin,β-catenin, c-ErbB2 and c-myc became apparently high from intestinal metaplasia and increased gradually in the evolutionary process of gastric adenocarcinoma. Significant differences were found among gastric adenocarcinoma and intestinal metaplasia according to the expressions of E-cadherin,β-catenin, c-ErbB2 and c-myc (P<0.05).Conclusions: Tissue microarray is a powerful tool for rapid identification of the... | | Keywords/Search Tags: | Tissue microarray, Immunohistochemical, Gastric adenocarcinoma, E-cadherin, β-catenin, c-ErbB2, c-myc | PDF Full Text Request | Related items |
