The traditional Chinese medicine, Yu ganzi(alias Amla,You ganzi,Niu ganguo), is dry fruit of plant phyllanthus emblica L.( Phyllanthus) of Euphorbiaceae. It has been used widely for anti-inflammatory and antipyretic drug in many local traditional medicinal systems, such as Chinese herbal medicine, Tibetan medicine and Ayurvedic medicine.Modern pharmacological studies indicate that the extract of Yu ganzi has protective effect for liver damnification, and preparation of Yu ganzi has been used in clinic. However, the base-substances for anti-HBV have not been elucidated yet. In order to find the constituents against HBV of Yu ganzi, our dissertation choose HepG2 2.2.15 cell as model and the secretive changes of HBsAg and HBeAg as estimate guide-line, aim to find effective constituents from Yu ganzi. The dry-fruit crushed, is extracted by 90% EtOH, then re-extracted with ethyl acetate, n-butanol successively to give three fractions, among of which, fraction ethyl acetate was the most effective one.Ten compounds were separated and determined from fraction ethyl acetate, including β-Amyrenone(I), Gallic acid(II), β-Daucosterol(III), β-Sitosterol(IV), Mucic acid-1-methyl ester-6-ethyl ester(V), Mucic acid dimethyl ester-2-O-gallate(VI), Ellagic acid (VII), Elaeocarpusin(VIII), 1,6-di-O-galloyl-β-D-glucose (IX), Digallic acid (X). Compound V was found from Yu ganzi for the first time, Compound I was found from the genus Phyllanthus for the first time.The compounds separated above were screened by HepG2 2.2.15 cell model, four of which(X, VIII, IX, II) showed remarkable anti-HBV activities. Compound X, VIII and IX showed strong inhibition for the secretion of HBeAg, IC50 were 5.5, 10.2and 13.2μg/ml respectively. Compound VIII, II and X showed inhibition in some extent for the secretion of HBsAg, IC50 were 70.9, 83.9 and 101.0μg/ml respectively. The anti-HBV activities of compound VIII,IX and X were indicated firstly.
