HPV Infection And Its Relationship With C-erbB-2 Over-Expression In Breast Cancer

[Objective ] :Oncogenic human papillomavirus (HPV)type 16 has been generally associated with a variety of epithelial cancers at different organ sites in human included breast. But the findings of HPV in breast carcinoma have remained controversial.[Methods] :61 breast cancer tissues, 27 normal adjacent breast cancer tissues, 29 benign breast tissues and 6 breast cancer cell lines' DNA were extracted and paraffin embedded sections were collected in this study. Using polymerase chain reaction (PCR) and in situ hybridization (ISH) methods, we have detected the existence and expression of HPV-16 E6, which has been considered as a major onco-protein with its ability to interact with p53.[Result] :The results have shown that: 65.6%(40/61) of breast cancer tissues, 65.2%(15/23) of benign tissues, 37.04%(10/27) of normal adjacent tissues tested by PCR was positive of HPV-16 E6 gene, 44.3%(27/61) of breast cancer tissues, 44.8%(13/29) of benign tissues was positive in E6 gene expression in cytoplasma shown by ISH, but none of normal adjacent tissues expressed E6 gene. Rate of consistence by two methods was 72.1%. In these 61 breast cancer tissues, 42.6%(26/61) expressed c-erbB-2 gene. In addition, 66.7%(18/27) of E6 gene expressing tumor samples were also positive in c-erbB-2 gene expression, and 76.5%(26-34) negative cases were also negative in c-erbB-2 gene.
