| Objectives To investigate the effects of FAK PCNA antisense oligonucleotides on the human stomach carcinoma cells via synthesizing FAK PCNA antisense oligonucleotides to work on human stomach carcinoma cells-SGC7901. Methods The antisense oligonucleotides were synthesized complementary to the protein encoding regions of FAKmRNA and PCNAmRNA and introduced into the human stomach carcinoma cells-SGC7901 respectively or associately. Theeffections were estimated by cell colony forming cell growth curves cellimmunochemistry staining method reverse transcription-Polymerase ChainReaction(RT-PCR) and Tunnel Method.Results The cell proliferation was inhibited effectively by antisenseoligonucleotides, the colony formation rate descend sharply, the inhibition effects tapered after 72 hours, the expressions of FAKmRNA PCNAmRNA and their protein also decreased sharply and the characteristic apoptosis was induced. Conclusion The FAKn PCNA antisense oligonucleotides can inhibits the human stomach carcinoma cells' proliferation and the expressions of FAKmRNA PCNAmRNA and their protein effectively.The antisense oligonucleotides can induce the human stomach carcinoma cells to apoptosis. |
