Objectives:To explore the feasibility of autologous skeletal muscle satellite cell transplanted periurethrally as a potential treatment of stress urinary incontinence (SUI),thereby to provide a new injection material for injection treatment of SUI.Methods:1.Primary rat skeletal muscle satellite cell was cultivated in vitro. The cultured cells were identified with cellular immunochemical stain. 2.The cells were incubated with Fluoro-Gold(a vital stain).The labeled cells were injected periurethrally into the autologous rats.3.The tissues were harvested 3-4 days after injection,sectioned,fixed.The slides were observed under fluorescence microscopy and examined immunohistochemically for desmin.Results:The primary skeletal muscle satellite cells were positive by cellular immunochemical stain with desmin. 100% cell nucleus were labeled,the color of fluorescence is inaurate. The survival rate of labeled cell was about 95%. Fluorescence microscopy of tissue sections from injected site can find fluorescently-labeled cell. Tissue sections stained positive for desmin.
