Its mechanism of inducing cancer cells apoptosis was also investigated.Firstly, the proliferation-inhibiting effect of HY-1 was detected by SRB assay on K562, HCT-15, A2780 and HT1080 cells in vitro. The results showed that HY-1 could respectively inhibited the proliferation of the four tumor cell lines with the IC50 values of 29.048±0.902μM for K562, 37.058 ±1.127μM for HCT-15, 30.351 ±0.597μM for A2780 and 31.491 ±0.095μM for HT1080. Moreover, the IC50 values of HY-1 is obviously less than that of the positive control cisplatin(CDDP).Secondly, the relatively sensitive cells K562 and relatively non-sensitive cells HCT-15 were selected from above four tumor cell lines to detect their cell-cycle distribution with PI single staining by FCM after treated with HY-1 in different dosage and time. The data indicated that the SubG1 peak percentage remarkably increased in dosage- and time-dependent way.
