Grifola frondosa, a highly regarded edible fungus, has many pharmaceutical activities, such as anticancer, antiviral and enhancing immunity of human body, on which polysaccharides purified from Grifola frondosa play a great important role. Large-scale fermentation has been available since the liquid-cultivation technique was applied which enabled the fermented mycelium the main source of GF polysaccharides.In this paper, a water-soluble polysaccharide fraction GF6F2 is extracted from fermented Grifola frondosa mycelia using a series of separation and purification methods. The primary structure of GF6F2 is determined using several chemical and instrumental analysis techniques. The structure-defined polysaccharide was further studied of its fundamental chemical & physical characters and stabilities.The fermented Grifola frondosa mycelium powder was extracted by ethanol, hot water, cool alkali and hot alkali. One water-soluble crude polysaccharide fraction GF6 was acquired (6.8% yield) and further purified by Ion Exchange Chromatography and Gel Filtration Chromatography method. A pure polysaccharide fraction GF6F2, which contained a little amount of protein, has been obtained.The primary structure of GF6F2 was elucidated by a series of chemical methods and instrumental techniques, such as TLC, GC, periodate oxidation, Smith degradation, methylation, acetolysis, partially acid hydrolysis and IR, UV, NMR, MS, GC-MS etc.. The results showed that GF6F2 has a relatively complex structure with aβ-1,3-Glc linked,β-1,6-Glc branched core structure and a 50% amount ofβ-1,3-/β-1,6-Glc branches through acid hydrolysis method.
