916 is one kind of polyanion sulfated amino polysaccharide which is derived from marine chitin and by ways of multiple molecular modification including deacetylation,carboxymethlation and sulfatation.After systematical research of pharmacology,pharmacodynamics and toxicology,916 shows good antiathero- sclerotic activity and was authorized to go into clinical study further by National Drug and Foods Administration at the name of 1 level new drug.The purpose of this paper is to affording prophase study of 916 in demic pharmacokinetics. A analytical method in biological sample-animal serum is established by HPLC. In this paper,post–column fluorescent derivation HPLC is used to analyze 916 in animal serum using guanidine hydrochloride, chromatography was carried out using a C8 reversed-phase column with an isocratic mobile phase consisting of methanol-water(20:80,v/v) at a flow rate of 0.2 ml/min. The postcolumn procedure involved fluorometric reaction with guanidine hydrochloride at 110℃. 916 was measured atλEx 249 nm andλEm 435 nm. 60% acetonitrile was selected to precipitate protein, the supernatant was submitted to the HPLC system and analyzed. Building standard curve and liner range, the limit of detection and limit of quantitation are determined. The precision, reproducibility and recovery of 916 in animal serum are tested.. This method provides a new simple, reproducible and validated assay, so it was successfully applied to the pharmacokinetic study of 916 in animals.In this paper, post-column fluorescent derivation RP-HPLC is firstly used to analyzed916 in animal serum. This method is used to detect 916 in rabbit serum after i.v. and in rat serum after oral administration. Serum concentration profiles are built.
