Objective: To construct dermal scaffold consist of collagen sponge then foster three-degrees artificial skin about epidermis and demis.Methods: First we cultivate keratinocytes and fibroblasts by routine techniques of cells cultivation, put collagen solution in—70 ℃ refrigeratory, then manufactured by freeze-drying collagen and cross-linked with ultraviolet and 0.25%glutaraldehyde. Second, to seed keratinocytes and fibroblasts at the bilayered scaffold separately and obtain the three-degrees artificial skin by cultivation.Result: Collagen scaffold gained the fittest pore size to cell growth at a freezing temperature of — 70 ℃. The two methods of crosslink improved mechanical property and increased the resistance of collagen membranes to enzymatic digestion.Conclusion: Collagen is a favorable material as dermal scaffold. The pore size of collagen scaffold was found to be dependent on freezing temperature, and was optimized at — 70 ℃. The mechanical property and the resistance to enzymatic digestion of collagen sponge were improved by ultraviolet and glutaraldehyde crosslinking, moreover, crosslink was impact hardly on the pore size of collagen sponge.
