Objective: 1 .To investigate the expression of CX/GJIC during different stages of AHF in rat. 2. To explore the effect of the down-regulate of CX/GJIC in repair of AHF. 3. If PNS has any influence in repair of AHF and its mechanism.Methods: Male Wistar rat 260 weighing 180-200g are given suitability feeding at first for a week, then random draw 60 from it as phenol barbital group (group PB, n=60) feeded with 800PPM phenol barbital for a week. The rest random divided into three groups: Panax notoginseng saponins group (group PNS, n=70), model group (group Model, n=100) and control group (group Control, n=30). The three groups besides group Control are intraperitoneal injected mixture of CCL4 and olive oil (V: V=l :1) only one time to make model. Three days before modeling all the animals (including the group Control) are kept on intragastric administration with gentamycin and lactulose till three days after modeling. After modeling, all the animals alive are taken specimens by opening abdomen separately in 1, 3, 7,10 and 14 days for blood biochemistry, pathohistology, immunohistolgy, Incision Loading/Dye-Transfer (IL/DT) and RT-PCR to detect the relationship between the degree of injury in different stages, the level and function of the CX/GJIC and the relation between the PNS and the repair of hepatic injury.Result: 1.after modeling all the animals show up depressed, activity and ingestion decreased lethargy, color pattern gloomy, abdominal distention, urine color yellow, action step down, severity case accompany with multi-organ hemorrhage, hyperspasmia, coma even death. The differential mortality between group PB and Model is significant (P<0.01), demonstrate that down-regulate CX/GJIC can degrade the mortality of AHF animal. 2.
