| Objective: To confirm the existence of mesenchymal stem cells in the umbilical cord blood;To compare the immunomodulatory functions of UCB-MSCs and human BM-MSCs in the experiment of PHA induction proliferation; To explore thedose-effect relationship of immunomodulatory functions of UCB-MSCs;To determine the lever of transforming factor betal(TGF-β1) of different groups, and analyze the relationship between immunomodulatory functions of UCB-MSCs and TGF-β1;To determine the effect of heparin on the immunomodulatory functions of UCB-MSCs and the lever of TGF-β1. To provide the guiding of using UCB-MSCs in clinical application.Methods: In intro UCB-MSCs and BM-MSCs were generated, their Morphology, phenotypes and Functions were investigated; UCB-MSCs and BM-MSCs cultured with lymphycytes in experiment induced by PHA , the proliferation of lymphocytes was measured by MTT, effects of UCB-MSCs and BM-MSCs on PHA-induced lymophocyte proliferation were investigated;The lever of TGF-β1 secreted by UCB-MSCs and BM-MSCs were determined by ELISA, the relationship between the effect of MSCs on the lymphocyte proliferation and TGF-β1 was analyzed; effects of heparin on the immunomodulatory functions of UCB-MSCs and the lever of TGF-β1 were investigated using the methods above.Result: (1) In intro UCB-MSCs and BM-MSCs were generated successfully.
