Effects Of Shu Gan Jie Yu Yin On Behavior, The Level Of CORT And ACTH In Plasma And The Level Of CRH In Thalamic In Depressed Model Rats

Depression is a disease of affective disorder. At the begining, the symptom is ademosyne, pessimistic emotion, Suspiciousness, etc, which is called depressive state.It may appear poor appetite, dyspepsia, insomnia and dreamful sleep, cardiopalmus and short breath, dizziness which belong to Functional disorder of organs If not cure. That is the Depression. The morbidity of depression increased fast year by year, because the rhythm of life is fast and mental stress has become much stronger than before.The reports about Pathogenesis of depression consider that it maybe have the association with monoamine neurotransmitter﹑neurobiochemistry﹑ endocrine system﹑ cardiovascular system﹑genetic factor and social psychic factor, etc. As one part of the Pathogenesis of depression research, this experiment is focuse on the relations between hormone level of Hypothalamus- Pituitary-Adrenal axis and depressive disorder. HPA axis is the most significant endocrine regulation system of the body. when stimulus affect on organism, the cerebral cortex send out information to amygdaloid body to cause CRH secrete. ACTH in blood which was secreted by hypophysis because of the CRH, can cause the adrenal cortex liberate adrenal cortex hormone. Hydrocortisone is one of the most important adrenal cortex hormone. HPA axis in human body can protecte andadapt the external irritant, therefore, CRH, ACTH, and hydrocortisone increase in patients'serum of the depressive disorder.Therapeutics of the depression experienced three stages: monoamine oxidase inhibitor (MAOI); tricyclic antidepressants; serotonin retake inhibitor. It is recognized that liver-energy has the most close relationship with the desease. Because liver controlling dispersion, if the function of liver is normal, then the functional activities of vital energy willl ascending-descending and coming in-going out and the physiologic function of entrails will also be normal. One week is a cycle. Afrer 28days, rats were randomly divided into six groups: control group, depression model group, low, middle and high dose SGJYY groups and Fluoxetine group. the control group drench the distilled water and depression model group drench the physiological saline. The cure keep 28days. During this period, The stimuli still continu. At the same time, we judge the degree of depression of rats by observing and recording the body weight and behavior moment times of rats, and detecting the ethology of the rats by"sacchar-consumption test"and"the experiment of box and square". At 56th day, we put the rats to death. Then we determine the content of CORT and ACTH in plasma which is separated from the blood of eyeball by radio-immunity and determine the CRH in hypothalamus in the same way. All the dates were dealed with statistics.RESULT: Compared with control group, the increase of body wight of depression model group is step down obviously. The times of horizontal movement and vertical movement all decreased remarkedly. The reduction sucrose solution consumption is obviously. The contents of CORT, ACTH and CRH are increase obviousely. But thebody wight and horizontal movement and vertical movement and sucrose solution consumption of SGJYY groups and Fluoxetine group all increased obviously, and the contents of CROT, ACTH and CRT decreased compared with the depression model group.CONCLUSION: chronicity stress could cause the body weight loss,decreased of activities,food consumption and accentuation of HPA of experimental rats. SGJYY could improve the behavioral items of the depression rats. It can decreased the level of CORT,ACTH in serum and the over-secrete of CRH and adjusted the functional disorder of HPAA accordingly.
