The diabetes mellitus is a kind of very ancient disease, the Chinese medicine calls it as "wasting-thirst ", namely emaciation and polydipsia, the modern medical science discovers that it is a kind of familiar metabolism disease.Because the change of the people living standard, working pattern and food and drink construction; as well as the average duration of life extend, the outbreak rate of diabetes mellitus increases high year by year.Because the diabetes mellitus is jeopardize to the human body only next to cancer, so it is called the second killhand of the modern disease.In fact, the diabetes mellitus is not fear, but terrible of it is complications.The harm which the diabetes mellitus brings, is almost come from its complications.The result of the genetics etiopathogenisis research is indicate the diabetes mellitus is a kind of polygene hereditary diseases in recent years, in it great vessels and capillary vessel which to be involved in organ, the heart and kidney pathological changes are the most serious complications, and is a diabetically the main reason to die, and may be like other complications, are subjected to the patient's heredity background influence. Therefore looking for and establishing effectively onset risk and preonset diagnosis forecast technique for cardiovascular diagnose and kidney neopathy , in clinical will have importance practical value.So the research of the related gene seems to be importance, scholars in all countries all pay tremendous close attention to this, but the indeed etiopathogenesis is still not clear.The experiment objective is to investigate whether ACE gene I/D polymorphism is a predictor of onset and progression of diabetic mellitus is incorporate nephropathy or Coronary heart disease of Chinese person in Jilin region.The experiment methods are that 1,the first step is subdivided 40 Chinese of Jilin reigon patients with diabetic coronary heart disease as the first group , 40 Chinese of Jilin reigon patients with diabetic nephropathy as the second group , 30 Chinese of Jilin reigon patients with diabetes mellitus as the third group, 30 Chinese of Jilin reigon health person contrast control as the fourth group. Then take diabetic coronary heart disease group, diabetes mellitus group and health person contrast control group act as the first part, carry on ACE gene I/D polymorphism contrast control study. Take diabetic nephropathy group, diabetes mellitus group and health person contrast control group act as the second part ,carry on ACE gene I/D polymorphism contrast control study. 2,The second step is abstracting template DNA, adopt the American Promega company produce of the DNA extract kit quickly abstracting person`s white cell DNA of peripheral blood. 3,the third step is the ACE gene expand the 16th intron of 287 bp insertion/deletion polymorphism was determind by polymerase chain reaction (PCR). 4,the fourth step is 1.5% agarose gel electrophoresis was performed,the results were observed under ultraviolet light. Confirm each genotype, compute ACE gene genotypic frequency and allelomorph frequency of ACE gene . make use of Hardy-Weinburg inherit quilibrium laws examine whether each genotype frequency attains heredity balance or not and have a community representative.Make use of chi-square criterion to compare allelomorph and genetype frequency and association between ACE gene insertion/deletion polymorphism and Type 2 diabetes with nephropathy or coronary heart disease in all groups .The experiment results are that 1,Hardy-Weinburg inherit quilibrium laws examination result is that each genotype frequency attains heredity balance and have a community representative . 2,in the second part , each genotype frequency of ACE gene respectively are that II type is 33%, the ID type is 45%, and the DD type is 23% of diabetic nephropathy group;each genotype frequency of ACE gene respectively are that II type is 30%, the ID type is 40%, and the DD type is 30% of diabetes mellitus group ;each genotype frequency of ACE gene respectively are that II type is 33%, the ID type is 43%, and the DD type is 23% of health person contrast control group. Be discovered by chi-square criterion,the ID genetype frequency and the I allelomorph frequency were higher in all groups;and the II genetype frequency was higher than the DD genetype frequency in all groups too. There was no significant difference between ACE gene frequency of genetype and allelomorph in these groups. 3,in the first part , each genotype frequency of ACE gene respectively are that II type is 13%, the ID type is 33%, and the DD type is 55% of diabetic coronary heart disease group;each genotype frequency of ACE gene respectively are that II type is 30%, the ID type is 40%, and the DD type is 30% of diabetes mellitus group ; each genotype frequency of ACE gene respectively are that II type is 33%, the ID type is 43%, and the DD type is 23% of health person contrast control group. Be discovered by chi-square criterion, in the diabetic coronary heart disease group the DD genetype frequency and the D allelomorph frequency are higher.Genetype frequency and allelomorph frequency of ACE gene distribute obvious difference of the other two groups.The experiment conclusion is that, the ID genetype frequency and the I allelomorph frequency were higher in diabetic nephropathy group , diabetes mellitus group and health person contrast control group . the statistics results show there are no significant difference between ACE gene frequency of genetype and allelomorph in these groups. But in the diabetic coronary heart disease group the DD genetype frequency and the D allelomorph frequency are higher.So ACE gene I/D polymorphism is a risk factor for Type 2 diabetic mellitus with coronary heart disease but is not associated with the development of Type 2 diabetic nephropathy among Chinese in Jilin reigon.
