Objective:To explore and develop the red blood membrane magnetic beads for ABO reverse grouping instead for fresh red cell and put it into auto reverse grouping system.Method:According to the reference about the synthesis of nano magnetic beads(MB),design the reasonable project.Test the irion content of the MB using ICP.Identify the chemical component of MB using IR and XRD.Anlysis the size using TEM ,and then determine the project of making MB.Prepare the red cell membrane and test the antigenicity and specificity using agglutination inhibition test.Research the combine method between cell membrane and MB.Evaluate the quality of the reverse reagent by comparing the specificity between flesh red cell and membrane magnetic beads.Result: Using co-precipitation method to make MB,the iron content is about 70% ,the main chemical component is Fe3O4,the size is about 200nm,it can be preserved in DW.The antigen of red cell membrane can inhibited antibody which titre is 64.Apply the reverse reagent to the auto blood typing system,testing 640 samples,the accuracy is 98.3%.Moreover test about 1500 samples that from clinic,the accurate is about 97.2%.Conculsion:The reverse blood grouping reagent prepared by nano magnetic beads can be used in clinic blood typing.Combining with auto system it can be used for large samples typing of ABO blood system.
