Background and Objective: Endometriosis (EM) has confirmed to be a"benign disease"with potential malignant, which was usually found in ovarian and the atypical endometriosis was thought to be the important foundation of this malignant process. Anti-oncogenes have the close correlation with most malignant tumors, which can suppress cell proliferation, prevent cell propagation and prevent malignant transformation. Pten has the max mutation rate in endometrioid adenocarcinoma, it's mutation had been found in ovarian cancer, which transformed from endometriosis. Pten protein will be the significance level in malignant transformation of ovarian endometriosis. P16 regulates cell cycle negatively and suppress cell proliferation, prevent cell propagation and prevent malignant transformation. The absence or mutation of p16 had been found in most malignant tumors. Fhit act in cell cycle and promote cell apoptosis, the decrease of expresstion of fhit protein has close correlation with endometrial cancer. There has not seen any report about p16, fhit in malignant transformation of ovarian endometriosis. We detected the expressions of PTEN, P16, FHIT proteins to find their impacts in malignant transformation of ovarian endometriosis.Methods: Immunohistochemical technique(s-p) was used to detect the PTEN,P16,FHIT protein expressions in endometriosis (Groupâ… ,26patients), atypical hyperplasia of ectopic ovarian endometrial glands (Groupâ…¡,14patients) and endometriosis with malignant change (Groupâ…¢,29patients).Results: 1.The expression of PTEN protein mostly located in cell nucelus,the positive cell distributed dispersively or locally. The positive rates of PTEN protein expression were 38% and 31% in Groupâ… and Groupâ…¢, andthere was no significant differenc(ep>0.05). The positive rate of PTEN protein (82%) in Groupâ…¡was significantly higher than that in Groupâ… and Groupâ…¢(p<0.05).2. The expression of P16 protein mostly located in cell plasma, the positive cell distributed dispersively or locally. The positive rates of P16 protein expression were 71% and 69% in Groupâ…¡and Groupâ…¢, and there was no significant difference(p>0.05). The positive rate of P16 protein (35%) in Groupâ… was significantly lower than that in Groupâ…¡and Groupâ…¢(p<0.05).3. The expression of FHIT protein was located in cell plasma and nucleus, the positive cell distributed dispersively or locally. The expression can be seen in all the three kinds of tissue and the positive rate were 88%,92% and 93%,respectively, which trend was upgraded .but has no significant different with each other(p>0.05).Conclusion: 1. The positive rate of PTEN protein in atypical hyperplasia of ectopic ovarian endometrial glands was significantly higher than those in ovarian endometriosis and carcinoma. And the positive rate in endometriosis was slightly lower than that in carcinoma. The express of PTEN may be the morning incident in malignant transformation of ovarian endometriosis.2. The positive rate of P16 protein in atypical hyperplasia of ectopic ovarian endometrial glands and endometriosis with malignant change were significantly higher than that in ovarian endometriosis. And the positive rate in endometriosis with malignant change was slightly lower than that in atypical hyperplasia of ectopic ovarian endometrial glands. Maybe there is another anti-oncogene(Rb) which would help P16 to act in malignant transformation of ovarian endometriosis and played an important role in this neoplastic process.3. The positive rates of FHIT protein upgraded in these three tissues. FHIT may be one of the factors in malignant transformation of ovarian endometriosis.
