Objective:Diabetes mellitus(DM) is a syndrome made up of chronic high glucose and complication with obvious abnormality,which is aroused by genetic, environmental and immune factors.Morbidity of DM is increasing of late years,the global patients were alreay excessed 194 millions in the report of the IDF in 2003,the number will increase a double in 2025(333 millions).The type 2 diabetes is more than 85%-95% in DM in abroad,and more than 98% in inland.Therefore,the study of Establishment of the animal model of type 2 diabetes have important medical significance.This study will filtrate the best animals, times,doses and establish test project by comparing examine tagets between rats and mouse.Methods:1. Choose 40 SD rats (180-200g), divide into 3 groups at random.The normal group (10) are raised by common forage,the fatness group(10) and experiment group(20) are raised by high fat forage.The experiment group rats are given 40mg/kg streptozocin(STZ) through intraperitoneal injection(ip).The rats are determined DM which value of fasting plasma glucode reach 16.7mmol·L-1 . Blood is taken out by picking eyeball for measure FPG,TC,TG and serum insulin.liver,kidney,pancreatic,Thymus and Spleen of all rats are taken out for later work.2. Choose 30 ICR mouse (18-20g), divide into 3 groups at random.The normal group (10) are raised by common forage,the fatness group(10) andexperiment group(10) are raised by high fat forage.The experiment group mouse are given 100mg/kg streptozocin(STZ) through intraperitoneal injection(ip).The mouse are determined DM which value of fasting plasma glucode reach 11.1mmol·L-1 . Blood is taken out by picking eyeball for measure FPG,TC,TG and serum insulin.liver,kidney,pancreatic and Spleen of all rats are taken out for later work.Resultes:1. After 28 days of raised high fat forage,TCand LDL level of the HFF group rats are higher than the negative control group(p<0.01),TG level higher than the normal group(p<0.05);After 21 days of raised high fat forage,TC level of the HFF group mouse are higher than NC group,but there are no statistical differences on TG(p>0.05).2. On the 7th day and 14th day of STZ injection,level of FPG of experiment group rats increase significantly compared with NC group and HFF group(p<0.01);Level of FPG of experiment group mouse are higher than NC group and HFF group(p<0.05).3. During 7 days of STZ injection,food intake of experiment group rats have no statistical differences(p>0.05)and water intake increase significantly (p<0.01) and body weight decrease compared with NC group and HFF group(p<0.05); During 7 days of STZ injection, food intake of experiment group rats have no statistical differences(p>0.05), water intake increase compared with NC group and HFF group(p<0.05),body weight of HFF group increase compared with NC group and experiment group; During 14 days of STZ injection,food intake of experiment increase compared HFF group(p<0.05), water intake increase significantly(p<0.01); at the end of the experiment ,the level of serum insulin have no statistical differences compared all groups(p>0.05).4. After 14 days of STZ injection,liver index and kidney index of experiment group rats are higher than NC group and HFF group(p<0.01),and Thymus index is lower than NC group and HFF group(p<0.05),there are no statistical differences compared among spleen index and pancreas index of all groups(p>0.05);liver index of experiment mouse is higher than NC group and HFF group(p<0.05),there are no statistical differences compared among spleen ,index,kidney index and pancreas index(p.0.05).Conclusion:1. Stable diabetes mellitus modle with hyperlipemia and insulin resist can been acquired after SD rats are given 40mg·kg-1 STZ and high fat forage 28days; Stable diabetes mellitus modle with hyperlipemia and insulin resist can been acquired after ICR mouses are given 100mg·kg-1 STZ and high fat forage 21days.2. Effect of STZ on pancreasβcells is definite demage.3. STZ cooperate with high fat forage can chang the index of liver,kidney and Thymus.4. Establishment of the animal model of diabete using rats are better than mouse by synthesis analysing.
