| Reconstruction of tissue-engineered uterus using typeâ… collagen gelUterus is an important reproductive organ of female. It is the birthplace of both menstruation and fetal. Additionally, uterus also has other important physiological functions. Thus, reconstruction of uterus in vitro is useful in studying the function of uterus and treating uterine diseases. Tissue-engineering is a hopeful way in the reconstruction of uterus. More and more researchers are trying to construct tissue-engineered uterus in vitro. However, until now, there's still nobody made a perfect one.There are four parts in this research. Firstly, the effect of sex sterols on the endometrium in vivo was studied, and we tried to investigate the location of putative uterus stem cells by the identification of labeling retaining cells (LRC) in rabbit endometrium. Secondly, we studied the effect of sex sterols on the proliferation of endometrial cells in vitro. Thirdly, using typeâ… collagen gel and cultured endometrial cells, we reconstructed tissue-engineered endometrium in vitro, and the support effect on the mouse 1-cell embryos was studied to confirm the function of the tissue-engineered endometrium and cultured endometrial cells. Finally, we reconstructed the engineered uterus tissues (EUTs) which composed of engineered endometrium tissue and engineered uterus smooth muscle layer which under the engineered endometrium layer. The EUT was an ideal model in studying the earlier event of embryo development and implantation.
