OBJECTIVE This study is aimed to reseach the protective effect of safflower injection against hepatic ischemia reperfusion injury in rabbits and its possible mechanism.METHODS 30 adult and health rabbits were divided randomly into the following 3 groups (10 rabbits each group): Group A: normal control; Group B: ischemic reperfusion model; Group C: pretreated the ischemic reperfusion model with safflower injection (2.0ml/kg). All the medicines were injected from the otic vena at 20min before ischemia.The ischemic repersfuion model was made with Pringle's method by blocking the hepatic artery and portal vein with a clamp for 30 minutes, and then reperfusing for 120min. Blood samples were obtained to measure the changes of TNF-a and ALT, AST at 30min after ischemia and 120min after reperfusion. Liver tissues were collected at the same time to measure the changes of MDA, SOD and to observe histologic changes.RESULTS1. The blood resum's TNF-a, ALT, AST and the activity of MDA in Group B and Group C were all higher than that in Group A at 30min after ischemia and at 120min after reperfusion, while at the same time the SOD's activity of Group B and Group C were lower than Group A. And the injuries of liver were found by pathological examination. (These changes above all accorded with P<0.05 )2. The TNF-a, ALT, AST and the MDA in Group C were lower than that in Group B at these two time-points, while the SOD in Group C was higher than Group B. And the liver injuries of histological were obvious improvement. (These changes above all accorded with P<0.05)CONCLUSION1.The effect of safflower injection can decrease the level of TNF-a and reduce the hepatic ischemia reperfusion injury.2. Safflower injection can reduce the activity of MDA and improve the activity of SOD effectively, thus it can provent the hepatic ischemia reperfusion injury from the hurt of free radicals.
