Objective: To investigate the changes of aldose reductase(AR) activity inSHR and the intervention effect of captopril and eucommia lignin on ARactivity, and to provide experimental evidence for study on the effect andmechanism of aldose reductase on attenuatting hypertension-inducedcardiovascular remodeling.Methods: To observe whether the fluorescent value was affected byhaemoglobin, spotting interval, the time of aqueous bath arousing fluorescenceand storage time of NADP solution. It was optimized that theNADP-generating fluorimetry assayed erythrocytes AR activity. AR fromSHR(10, 14, 18w) erythrocytes were determined as improved previously.Ultraviolet spectrophotometry was performed to determine the activity of ARfrom cardiac muscle of 18w SHR that had been induced respectively byintragastric administration of normal saline, captopril and eucommia lignin.Absorption spectrometry and immumofluorescence method were performed todetermine the content of Blood plasm active mass, such as NO, RA, AngⅡ,CGRP.Results: (1)The Precision of NADP-generating fluorimetry was affectedby haemoglobin, spotting interval and the time of aqueous bath arousingfluorescence. NADP solution can be stored at -20℃over 4 week. (2)Therewas no evidently difference on AR activities of 10, 14w SHR erythrocytesthan control group(P>0.05). AR activities of 18w SHR erythrocytes(29.6±1.6) were significantly increased than control group (26.5±0.8)(P<0.05). AR activities of 18w SHR cardiac muscle (0.091±0.010) weresignificantly increased than control group (0.030±0.014) (P<0.05). (3)Theactivities of AR from cardiac muscle of 18w SHR that had been induced byintragastric administration of captopril (0.032±0.012) were significantlylower than by intragastric administration of normal saline(P<0.05), and wereno evidently difference than control group (P>0.05) The activities of AR from cardiac muscle of 18w SHR that had been induced by intragastricadministration of eucommia lignin (0.060±0.011) were significantly lowerthan by intragastric administration of normal saline(P<0.05), and were higherthan control group(P<0.05). The changed tendency of AR activity were notcorrelated with content of NO, RA, CGRP, were correlated with content ofAngⅡ(R=0.695, P=0.004).Conclusion: (1)NADP-generating fluorimetry that had been optimizedcan be accurately measured activity of erythrocyte AR. (2)SHR AR activitywere significantly increased than control group. (3)Captopril and eucommialignin can inhibit AR activity. The mechanism may be related to decreasingcontent of AngⅡ.
