Objective: To study the expression of substance p in rabbit anaphylactic shock and explore its contribution to anaphylactic shock, trying to seek objective morphological reference support to diagnose sudden erethistic death for forensic medicine.Methods: 30 rabbits were divided into two groups randomly, the experimental group which contains 24 rabbits and the control group which contains 6 rabbits. Rabbits in experimental group were made into anaphylacatic shock modes with human sera. Plug, stomach, colon, sacculus rotundus were extracted in different shock interval, including sudden death instantly, shock 0.5 hour, shock 1 hour, shock 2 hours.(1) Frozen section, routine HE staining. (2) SP were stained using immunohistochemistry(IHC) method, and the images of SP were analyzed by BI-2000,and the integral optical density (IOD) were calculated. (3) SP were stained using Hybridization in situ (ISH) method, and the images of SP were analyzed by BI-2000,and the IOD were calculated.Results: (1)IHC results: the protein expression of SP of the experimental groups are higher than that of the control group(P<0.001) and to waken gradually following the shock interval. Statistic ANOVA results are: stomach mucosa group F=198.008,colon mucosa group F=56.796, sacculus rotundus mucosa group F=67.231.SNK test results are: There are conspicuous differences between every groups of stomach. There are no statistically significant between the control group and shock 2 hours group of colon and sacculus rotundus. There are conspicuous differences between other groups of colon and sacculus rotundus. (2) ISH results: the mRNA expression of SP of the experimental groups are higher than that of the control group(P< 0.001) and to waken gradually following the shock interval. Statistic ANOVA results are: stomach mucosa group F=67.506,colon mucosa group F=72.117, sacculus rotundus mucosa group F=216.798.SNK test results are: There are conspicuous differences between every groups of colon, sacculus rotundus and stomach which has no statistically significant between the control group and shock 2 hours group .Conclusion: The protein and mRNA expression of Substance P was significantly stronger in gastrointestinal tissue and sacculus rotundus of rabbit anaphylactic shock. The expression of SP was to weaken gradually following the shock interval. It can be as an objective morphological reference support for diagnosing sudden erethistic death in forensic medicine.
