Objective To establish anaphylactic shock model induced by Ovalbumin(OVA) in mice and to study the space-time courses of MCT and ECP in Larynx, Lungs and Stomach after anaphylactic shock for diagnosing anaphylactic shock in the field of forensic and clinical medicine.Method (1) Fifty mice were divided into two groups: sensitized and control, the sensitized were immunized by intraperitoneal injection of OVA at first day and then duplicated after a week. Challenge was given by vena caudalis injection three weeks later. Before challenged, the sensitized were injected by intraperitoneal injection of Hanks solution and killed after an hour. Extracting peritoneal fluid and to calculate percentage of mast cell degranulation by microscope. (2) The sensitized of another fifty mice were killed after sensitization (the interval is 30min, 1h, 6h and 24h). After anatomy, Larynx, Lungs and Stomach were fixed, dehydrated for frozen section. Hybridization in situ and Image-Pro Plus5.0 were used to detective MCT and ECP.Result (1) Symptoms of anaphylactic shock appeared in sensitized group (shock rate 100%, death rate 22.5%). The percentage of mast cell degranulation was increased in sensitized group significantly. (2) MCT and ECP mRNA of Larynx, Lungs and Stomach were increased in sensitized group and the expressing peaks of MCTmRNA were found at 30min after sensitization, and then decreased. The expressing peak of ECP mRNA in Larynx was found at 30min and kept on high level. However, the peak of ECP mRNA in Lungs was found at 6h, 24h in stomach.Conclusions (1) An animal model of anaphylactic shock was established successfully and it proved high sensitivity to OVA in mice. (2) MCT and ECP mRNA in Larynx, Lungs and Stomach showed some regularity with space-time courses after anaphylactic shock in mice. MCT and ECP played roles in immediate reaction and late phase reaction and can provide objective diadynamic criteria to diagnose anaphylactic shock for forensic and clinical practice.
