Objective To investigate the mechanism of action of cytokine and nuclear factor in normal rats and diabetic rats following limbs ischemia reperfusion and the mechanism of protect action of propofol,the experiment was made on the changes of NF-κB and TNF-αin lung of diabetic rats following limbs ischemic reperfusion and the effect ofpropofol in the process.Method Adult male Sprague-Dawley rats,weighing 220-250g were randomized assigned to normal rats and diabetic rats,the normal rats were divided into three groups randomly:groupⅠA(n=6)control group,groupⅠB(n=6)limbs ischemic reperfusion group,groupⅠC(n=6)limbs ischemia reperfusion and propofol group;diabetic rats were divided into four groups randomly: groupⅡA(n=6)control group,groupⅡB(n=6)limbs ischemia reperfusion group,groupⅡC(n=6) propofol group,groupⅡD(n=6)limbs ischemia reperfusion and propofol group.The experiment deployed immunohistochemistry(IHC)in order to detect qualitatively and semiquantitatively the express of TNF-αand NF-κB and radio-immunity(RI)in order to detect quantitatively the level of TNF-α.Result First,limbs ischemic reperfusion(groupⅠB)caused lung damage compared with normal rat(groupⅠA),the level of NF-κB and TNF-αwas increased and there were statistical significance(P<0.05).Second,diabetic rats(groupⅡA)caused lung damage compared with normal rat(groupⅠA),the level of NF-κB and TNF-αwas increased and there were statistical significance(P<0.05).Third,diabetes mellitus with limb ischemia reperfusion caused lung significant damage,compared with normal rat(groupⅠA)and limbs ischemic reperfusion (groupⅠB)and diabetic rats(groupⅡA).the level of NF-κB and TNF-αwas predominantly high,and there were statistical significance(P<0.05).Fourth.compared with the intervention of propofol,every group level of NF-κB and TNF-αwas cut down after intervention of propofol and there were statistical significance(P<0.05).Fifth,there were not significant difference in the rest each other(P>0.05).Conclusion First,the level of NF-κB and TNF-αwas step up for diabetic rat and limb ischemia reperfusion rat and diabetes mellitus with limb ischemia reperfusion rat,moreover,the level is maximum for diabetes mellitus with limb ischemia reperfusion rat.Second.propofol can cut down the level of NF-κB and TNF-αfor diabetes mellitus rat and limb ischemia reperfusion rat's lung.It is to say that propofol with anti-inflammatory and anti-oxygen role has protection for the lung damage induced by limb ischemia reperfusion injury and diabetes mellitus injury.
