| Objective To extract polysaccharide from H.polybotrys Hand.-Mazz.( HPS) and investigate the growth inhibition and the results of apoptosis induced by HPS on MGC-803 cell line.Methods Crude water-soluble polysaccharides were isolated from the roots of HPS by hot water extraction followed by ethanolprecipitation.After removed starch and protein and fractionated by ultrafiltration and SephadexG-200 ,Its total soluble sugar content was detected colorimetrically by the phenol-sulfuric acid method. MTT assay was used to detect cell growth inhibition of HPS on MGC-803 cell line, fluorescence microscopy, electron microscopy, flow cytometry (FCM) were used to demonstrate the presence of apoptosis.Results1. HPS was found to be a single sugar ,The soluble sugar contents of HPS is 94.88%, RSD=1.05%.2. The experiment showed that 1.0-4.0mg/ml HPS could inhibit the growth of MGC-803 cells significantly in a dose and time dependent manner(P<0.05,compared with the matched control group)., and morphological characteristics of apoptosis was observed with condensation of nucleus, bubble of cytoplasm, fragment of nucleus.3. Sub-Gi peak and increased G0/G1 period cells were shown byFCM(P<0.05, compared with the matched control group).Conclusion1.HPS could induce the inhibition of MGC-803 cell growth by means of apoptosis, and the blocking of G0/G1 period cells is involved in the mechanism of apoptosis.2.the phenol-sulfuric acid method was simple,raprid and accurate in detecting the soluble sugar contents of HPS. |
